چکیده

The plant pathogen basidiomycete S. rolfsii secretes two laccases (SRL1 and SRL2) with molecular weights of 55 and 86 kDa, respectively. Laccase production was shown to be inducible by the addition of 2,5-xylidine to the cultural media. After treatment with a combination of chitinase and -1,3-glucanase, two different laccases were isolated from the sclerotia depending on the stage of sclerotia development. The more prominent laccase, SRL1, was purified and found to decolourize the industrially important wool azo dye Diamond Black PV 200 without the addition of redox mediators. The enzyme (pI 5.2) was active in the acidic pH range, showing an optimal activity at pH 2.4, with ABTS as substrate. The optimum temperature for activity was determined to be 62 ◦C. Enzyme stability studies revealed that SRL1 was notably stable at 18 ◦C and pH 4.5, retaining almost full activity after a week. Oxidation of tyrosine was not detectable under the reaction conditions but the enzyme did oxidize a variety of the usual laccase substrates. SRL1 was strongly inhibited by sodium azide and fluoride. Dye solutions decolourized with the immobilized laccase were successfully used for redyeing. © 2003 Elsevier Inc. All rights reserved.

Indigo degradation with purified laccases from Trametes hirsuta and Sclerotium rolfsii.

The degradation of the textile dye indigo with purified laccases from the fungi Trametes hirsuta (THL1 and THL2) and Sclerotium rolfsii (SRL1) was studied. All laccases were able to oxidize indigo yielding isatin (indole-2,3-dione), which was further decomposed to anthranilic acid (2-aminobenzoic acid). Based on the oxygen consumption rate of the laccases during indigo degradation, a potential ...

Pseudomonasextremorientalis BU118: a new salt-tolerant laccase-secreting bacterium with biotechnological potential in textile azo dye decolourization

The present investigation focused on screening of a new potent strain for laccase production and optimizing the process parameters to achieve the maximum enzymatic decolourization of textile azo dye Congo red. Seven hydrocarbonoclastic bacterial strains were selected as positive in laccase production in solid medium using 2,6 dimethoxyphenol as an enzyme activity indicator. The best enzyme prod...

Enhanced decolourization of the azo dye Sirius rose BB by laccase–HBT system

Response surface methodology (RSM) was applied to optimize the decolourization of the diazo dye Sirius rose BB (SR) by crude laccase from the white-rot fungus Trametes sp. strain CLBE55. A Box–Behnken design using RSM with six variables, namely pH, incubation temperature, enzyme (laccase) concentration, 1-hydroxybenzotriazol (HBT) concentration, dye concentration and incubation time was used in...

Remazol Brilliant Blue R Dye Decolourization by Laccase Produced by Pleurotus Sajor-caju via Solid-State Fermentation

The application of synthetic dyes in various industries improved the aesthetic properties of the products, but the discharge of dye-containing effluent into water bodies leads to serious environmental problems. Till date, colour removal from effluent is still a challenge due to the absence of economically attractive treatments. In this study, decolourization of a synthetic dye – Remazol brillia...

Isolation and characterization of glyoxylate dehydrogenase from the fungus Sclerotium rolfsii.

Glyoxylate dehydrogenase (glyoxylate:NAD+ oxidoreductase) was purified 600-fold in three steps from crude extracts of the fungus Sclerotium rolfsii (Corticium rolfsii Curzi). Two of the purification steps involved dye-affinity chromatography. The enzyme is a tetramer of Mr 250 000, with identical subunits of Mr 57 000. Inhibition studies suggest that there is one essential thiol group per activ...
